Abstract: The PCR technique was used to amplify the mtDNA D-Loop in 30 individuals of Paramisgurnus dabryanus collected from the Nansi Lake.The PCR products were purified and sequenced,and then 963bp nucleotide sequences of partial D-Loop were obtained.CLUSTAL X(1.83) to align and comparison of the sequences of these 30 individuals with each other revealed that there were 33 variation sites including 12 transition sites and 21 transversion sites.The pairwise genetic distances were computed by MEGA,and the UPGMA and NJ phylogenetic trees was established through the cluster analysis over the pairwise genetic distance.DNASP showed that the number of haplotypes(h) was found to be 15,the haplotype diversity(H) 0.899,nucleotide diversity(Pi) 0.008 and the average number of nucleotide diferences(K) 7.79.It can be concluded that the variation in mtDNA D-Loop of Paramisgurnus dabryanus in the Nansi Lake is relatively rich and wealthy on genetic diversity.
